Background: Several single nucleotide polymorphisms (SNP) of adiponectin gene (AdipoQ) have been related to T2DM among which rs2241766(+45T>G) and rs1501299(+276G>T) are important ones.
Objective: To see the relation of rs2241766(+45T>G) and rs1501299(+276G>T) with young diabetic subjects.
Methods: This observational cross-sectional study was carried out in 62 DM and 57 age- matched NGT, aged 18-30 years. Anthropometry, BP, plasma glucose and lipid measurement and genotyping for rs2241766(+45T>G) and rs1501299(+276G>T) were done. Genotyping was done by PCR-RFLP.
Results: The minor allele frequencies for rs2241766(+45T>G) and rs1501299(+276G>T) were G (0.15) and T (0.17), respectively. The observed genotype frequencies of rs2241766(+45T>G) and rs1501299(+276G>T) maintained Hardy-Weinberg equilibrium in both study groups. There was no association of rs2241766(+45T>G) [recessive: OR=0.36 (95%CI:0.04-3.61), p=0.36] and rs1501299(+276G>T) [over-dominant: OR=0.54 (95%CI:0.24-1.21), p=0.13] with DM as well as no difference between DM and NGT in the estimated haplotype frequencies
of these two polymorphisms. Linkage-disequilibrium analysis showed the absence (r2=0.0012, p=0.6106) of linkage-disequilibrium between them. Clinical and biochemical parameters didn’t differ between those having a pair of the wild-type allele and those having at least one variant-type allele as well as among haplotypes estimated. When compared in a subgroup of subjects without insulin resistance, rs2241766(+45T>G) genotype was associated with DM [OR=4.773, p=0.034]. Logistic regression analysis also predicts that there is a 4.8 times higher likelihood of developing DM for those having a “G” allele in rs2241766(+45T>G) [OR=4.773, p=0.026] position in this subgroup.
Conclusions: SNP rs2241766(+45T>G) & rs1501299(+276G>T), neither independently nor as haplotype, increase the likelihood of developing DM in the young as well as not associated with metabolic syndrome. Although they don’t influence the risk of DM in general, they might increase the likelihood of developing DM in the young subjects without insulin resistant.
